To consume the SAP Business One Service Layer, I need to create an API where I will route to my destination.

if you define the B1 service layer as your destination -you should consume B1 Service Layer from the BTP.When configuring the B1 service layer as your destination on BTP, make sure to utilize basic authentication in your destination configuration.
